Go to the potion seller in any town, and select edit skill. There, for some gold, you can delevel skills and masteries. Remember that you cannot delete a skill/mastery until all skills are deleveled that require that skill/mastery.

After obtaining the potion, right click it. An interface should open with your skills there and you can increase/decrease your skills/masteries in that window.
